Illustrative diagram showing the pigmentation of the skin as it occurs from the deeper layers of the skin, up to the stratum corneum.
Test system: human keratinocyte
Antioxidant potential (ROS): Dichlorodihydro-fluorescein diacetate (DCFH-DA) assay
The test uses DCFH-DA, a cell permeant reagent fluorogenic dye. After cell uptake, DCFH-DA reacts with cell esterases to form DCFH, a non-fluorescent compound, which then oxidised by ROS to form DCF. Since DCF is fluorescent, it can be detected by fluorimeter, flow cytometry, or fluorescence microscopy.
